Prion disease is a group of infectious, chronic progressive and fatal spongiform encephalopathy caused by mutated prion protein. Prion disease was first found in animals. It was discovered 200 years later that prion disease can occur in human beings. There are four kinds of prion diseases in human and animal.

Its core is a 27-30kd protein, which is called prp27-30. The core of this thermostable protease is covered with a protein with a molecular weight of 33-35kd, called prp33-35 or PrPSc. Another protein similar to PrPSc, PrPc, is found in the brain of uninfected normal animals. It can tolerate protease digestion and polymerize into a rod-shaped structure called PrP rod or pruritus associated filaments (SAF).
